The distance between Sun City and Cape Town is about an hour by road. the big distance involved is between them and Cape Town. You are looking at a good 17 hour by road which is best broken up over 2 days. Where you want to overnight depends on budget and what you would like to see.
There are private charter flights that fly regularly between Madikwe and Cape Town or you could drive/ transfer down to Joburg and take a scheduled flight from Lanseria or OR Tambo airport.

Madikwe is a premier big 5 safari destination, with most of the lodges being either 5 or 4 star quality. The game reserve is famous for its Wild Dogs and is only 3 hours from Joburg
Sun City is famous for it Golf Courses, its shows, the waterpark with wave pool and of course its Casinos . You are never short of anything to do here. There are 4 different accommodation types to choose from here including the world famous "Lost City".
Inn on the Square is bang in the middle of Cape Town with easy access to all areas. Public Transport is well run in Cape Town and there are many transfer options for the beaches, Table Mountain and The Waterfront.
